摘要
[目的]探讨肺纤维化大鼠肺组织中瘦素的表达及意义.[方法]选择Sprague-Dawley雄性大鼠36只,随机分为实验组和对照组,每组18只,实验组应用博莱霉素气管内注入建立实验性大鼠肺纤维化模型,对照组注入生理盐水,采用HE染色、Masson三联染色检测肺组织病理变化,免疫组织化学染色、RT-PCR等方法检测其肺组织中瘦素蛋白和mRNA表达水平.[结果]实验组大鼠d7呈现典型的肺泡炎、d14纤维组织增生、d28稳定的肺纤维化;瘦素蛋白和瘦素mRNA在对照组肺组织表达较弱,在实验组d7表达增强,d14进一步增强,d28减弱,但仍强于对照组(P〈0.05).[结论]肺组织内瘦素表达的上调可能参与肺纤维化的发生和发展.
[Objective] To explore the expression of leptin in lung tissues of rats with lung fibrosis and its significance. [Methods] Thirty six male SD rats were randomly divided into experimental group and control group with 18 cases in each group. The experimental group received intratracheal injection of bleomycin for establishing rat model of experimental lung fibrosis. The control group was injected with saline. HE and Masson staining were used to detect pathological changes of the lung. Immunohistochemical staining and RT-PCR were used to detect the protein and mRNA expression of leptin in lung. [Results] Rats in experimental group showed typical alveolitis on dv , hyperplasia of fibrous tissue on d14 and stable pulmonary fibrosis on d2s. The protein and mRNA expression of leptin in lung of control group was weaker, but increased in the experimental group on d7 , and increased further on d14 , and weakened on d28, but still stronger than that in control group. [Conciusion] Upregulation of leptin expression in lung may be involved in the genesis and development of lung fibrosis.
