摘要
目的研究导痰汤是否能通过对细胞间黏附分子-1(ICAM-1)和JNK通路的调节而干预颈动脉粥样硬化。方法将25只雄性新西兰白兔随机分为对照组、模型组及导痰汤大、中、小剂量组。对照组给予基础饲料喂饲,模型组和导痰汤治疗组予以高脂饲料喂饲,并在喂饲1周后实施颈动脉内膜空气干燥术,术后治疗组分别予10、8、6 mL/kg导痰汤灌胃,每日给药1次。4周后处死家兔,取每只家兔右侧颈动脉远端1 cm,固定后行HE染色观察病理改变。应用免疫组化SABC法检测ICAM-1和JNK表达情况。结果模型组血管壁泡沫细胞、平滑肌细胞和内皮细胞的胞浆中有ICAM-1和JNK强阳性表达,明显高于对照组(P<0.01)。导痰汤治疗组细胞内ICAM-1和JNK阳性表达明显低于模型组(P<0.01),且随剂量的增加,对ICAM-1和JNK的抑制逐渐增强;JNK活性与ICAM-1水平呈显著正相关(r=0.848,P<0.01)。结论导痰汤可以有效抑制颈动脉硬化家兔血管内皮细胞中ICAM-1和JNK的表达。
Objective To research the influence of Daotan Decoction to the expression of ICAM-1 and JNK in carotid atherosclerosis rabbits.Methods Twenty-five male rabbits were divided into control group,model group and Daotan Decoction group(high-,medium-,low-dosage).Control group was treated with basic diet,model group and Daotan Decoction group were treated with both high-fat diet and carotid artery intima-air drying technique.Three dose of Daotan Decoction group was treated with Daotan Decoction 10,8,6 mL/kg respectively for 4 weeks.The right carotid was cut and fixed.The expression of ICAM-1 and JNK was observed by immunol histochemistry and pathological staining.Results Compared with control group,the expression of ICAM-1 and JNK in vascular foam cells,smooth muscle cells and endothelial cells of model group was significantly increased(P0.01).And compared with model group,Daotan Decoction could significantly restrain ICAM-1 and JNK expression(P0.05).Also,there was significant positive relationship between the expression of ICAM-1 and JNK(r=0.848,P0.01).Conclusion Daotan Decoction can restrain the expression of ICAM-1 and JNK in carotid atherosclerosis rabbits.
