高脂血症促进载脂蛋白E基因敲除小鼠牙周炎进展的实验研究 被引量:5

摘要 目的探讨高脂血症是否影响牙周炎进展。方法以ApoE基因敲除的C57BL/6J(ApoE-/-)小鼠和相同基因背景的野生型C57BL/6J(C57)小鼠各分为实验组和对照组,采用丝线结扎小鼠的上颌双侧第二磨牙,实验组接种牙龈卟啉单胞菌(P.gingivalis),对照组只完成牙周结扎过程而不接种细菌。用纸尖从牙周袋获取P.gingivalis进行菌落计数;截取上颌骨标本行亚甲基蓝染色,观察牙周组织损伤程度。结果普通饮食条件下,ApoE-/-小鼠血清脂质水平显著高于C57小鼠;实验第7d时实验组ApoE-/-小鼠牙周组织中获取的P.gingivalis菌落数为(8 341.66±1 217.88)个,显著高于C57小鼠(4 466.66±1 115.87)个(P<0.05);实验组ApoE-/-小鼠牙槽骨吸收量为(506.87±93.89)μm,显著高于C57小鼠(322.18±41.40)μm(P<0.05)。结论 ApoE-/-小鼠血清脂质水平升高,牙周清除P.gingivalis能力下降,牙周病进展加速。 Objective To explore the effect of hyperlipidemia on periodontitis progress.Methods Apolipoprotein E knockout C57BL6J(ApoE-/-) mice and the same genetic background wild type C57BL/6J(C57)mice were both divided randomly into experimental and control group,and mice in experimental group received periodontal ligation and Porphyromonas gingivalis(P.gingivalis) inoculation at bilateral maxillary 2nd molar.P.gingivalis was harvested from the crevice of the ligation teeth by paper point.Maxilla was collected for methylene blue staining to evaluate alveolar bone loss,and serum lipid level was evaluated.Results Under ordinary diet,serum lipid levels in ApoE-/-mice were significantly higher than those of C57 mice;On the 7th d of the experiment,P.gingivalis colony forming units harvested from the crevice of the ligation teeth were significantly increased in ApoE-/-mice(8 341.66±1 217.88) than those of C57 mice(4466.66±1115.87),and alveolar bone loss in ApoE-/-mice(506.87±93.89 μm) was significantly more serious than that of C57 mice(322.18±41.40 μm)(P0.05).Conclusion Serum lipid levels in ApoE-/-mice were increased,which could decelerate the ability to clear P.gingivalis in periodontal tissue and accelerat eriodontal disease progress.
