摘要
目的:构建我国8种重要病原真菌DNA条形码数据库,评价rDNA-ITS区在系统发育学中的价值方法:进行8种病原真菌的ITS区序列测定和序列分析,使用NJ法和BI法构建种系发育树。结果:ITS序列在2株须癣毛癣菌和2株红色毛癣菌之间完全一致;而在不同物种的真菌间,该序列存在数十到数百个核苷酸替代差异。依据ITS区构建的医学真菌系统发育树与传统分类学一致。结论:ITS区适合作为我国重要医学真菌物种鉴别的DNA条形码。
Objective: To construct medical fungi DNA barcoding for 8 important species, and to evaluate the phylogeny value of internal transcribed spacer of the nuclear ribosomal DNA in fungi. Method: ITS region of 8 med- ical fungi was sequenced and compared and phylogenetic trees were constructed using the neighbor - joining method and Bayesian Methods, separately. Results: ITS sequence was accordant in 2 strains of T. mentagrophytes and 2 strains of T. rubrum. Between species, the number of nucleotides was different from tens to hundreds. Phylogenetic trees of medical fungi based on ITS sequence were identical to conventional taxonomy results. Conclusion: ITS re- gion can be applicable to DNA barcode of our medical fungi identification.
