摘要 目的:评价马拉色菌随机扩增DNA多态性(RAPD)与生理生化方法在菌种分型上的差异及菌株DNA型别和菌种间的关系,分析其种间和种内的DNA多态性。方法:选取从马拉色菌毛囊炎分离出的马拉色菌,经生理生化及形态学方法鉴定的菌株52株,提取DNA,采用RAPD方法分析DNA多形性。结果:共有90个DNA片断被扩增,从RAPD带型画出的遗传树状图来看,RAPD与生理生化-形态学方法在菌种分型上存在差异,所有52个菌株均可见到一定程度的种间和种内DNA多态性。结论:二种方法在菌种分型上存在差异,菌株DNA型别和菌种间的关系不甚明确;种间、种内存在DNA多态性。 Objective: To evaluate the difference in species classification of Malassezia by using different (physi- ological- biochemical and morphological method, and random amplified polymorphic DNA (RAPD) analysis, and to determine the relationship between the genotypes and phenotypes of the Malassezia species, and to analyze the DNA polymorphism between inter- species and inter- strains of the fungi. Method: Malassezia DNA was extracted from 52 clinical strains obtained from patients with Malassezia folliculitis and typed by physiological - biochemical and morphological method. DNA was amplified and analyzed by RAPD method. Results: A total of 90 DNA fragments were amplified. There was polymorphism of Malassezia species between phenotypes and genotypes, according to the dendrogram based on the RAPD bands. Inter- species and inter- strains DNA polymorphism was presented in all the 52 Malassezia strainstested. Conclusion: There is typing difference of Malassezia species identified between phe- notype and genotype methods. The relationship between the types of DNA fragments and the species is not very clear. Both inter- species and inter- strains DNA polymorphism exist and the inter- strains polymorphism reflect better than the inter- species one.
