基因组高通量测序技术进展及其在预防医学中的应用 被引量:4

摘要 测序技术在生命科学研究中发挥着重要作用。研究人员借助第一代测序技术完成了人类基因组图谱的绘制,但其读取片段较长,一次仅读取数十条序列,导致测序结果的准确性受到影响,并且费时费力,因而很快被以高通量为主要特点的第二代测序技术所取代,其代表性技术包括美国罗氏公司的454技术、英国Illumina公司的Solexa技术和美国应用生物系统公司的SOLID技术。新涌现的第三代测序技术,以单分子实时(singlemoleculerealtime,SMRT)测序技术、Oxford的纳米孔测序技术和Helicos的基因分析系统为代表,与第二代测序技术相比,拥有更高的通量、准确度和更低的成本,
