摘要 目的探讨自噬相关基因Beclin1在鼻咽癌组织中的表达及其与临床病理特征的关系。方法 RT-PCR检测50例鼻咽癌、20例癌旁黏膜和15例鼻咽黏膜慢性炎症组织中Beclin1表达,并分析其与临床分期、病理组织类型和转移的关系。结果鼻咽癌、癌旁黏膜、慢性炎症组织中,Beclin1阳性表达率分别为48.0%、85.0%、93.3%,mRNA相对表达量分别为(0.658±0.158)、(0.802±0.086)、(0.809±0.097);鼻咽癌组织中Beclin1阳性表达率及mRNA相对表达量均显著低于癌旁黏膜及慢性炎症组织(P<0.05或0.01)。Beclin1 mRNA表达与鼻咽癌临床分型、组织学分型、淋巴结转移有关(P<0.01)。结论 Beclin1基因在鼻咽癌组织中表达下调,提示自噬活性的改变可能对鼻咽癌的发生、发展起一定作用。 Objective To study the relationship between expression of autophagy gene beclin1 and clinicopathologic features in nasopharyngeal carcinoma(NPC).Methods Beclin1 expression was detected by RT-PCR in 50 NPCs,20 paracancer mucosae and 15 chronic nasopharyngeal mucosal inflammations.The relationship between beclin1 mRNA expression and clinical staging,pathologic types,metastasis was analyzed.Results Beclin1 expression rates were 48.0%,85.0%,and 93.3%,and its mRNA levels were(0.658±0.158),(0.802±0.086),and(0.809±0.097) in NPC,paracancer mucosae and chronic inflammations,respectively.The positive expression rates and mRNA levels of beclin1 was significantly lower in NPC compared with paracancer mucosae and chronic inflammations(P0.05 or 0.01).Beclin1 mRNA expression was positively correlated with the clinical staging,pathological types,and neck lymph node metastasis in NPC(P0.01).Conclusion The downexpression of beclin1 gene suggests that the autophagy may play a role in development and progression of NPC.
