缺氧-复氧对肾小管上皮细胞内DRP1及OPA1表达的影响 被引量:3

Effect of hypoxia/reoxygenation on the expressions of DRP1 and OPA1 in epithelial cells of kidney tubules

摘要 目的:探讨缺氧-复氧(hypoxia/reoxygenation,H/R)后人近曲肾小管上皮细胞(HK-2)内动力相关蛋白1(Dynamin related protein 1,DRP1)及视神经萎缩症蛋白1(optic atrophy,OPA1)表达的变化。方法:以人近曲肾小管上皮细胞株为研究对象,将培养细胞随机分为正常对照组和H/R组。正常对照组常规培养,H/R组先缺氧24h,然后复氧培养6h。光镜观察细胞形态变化,CCK-8检测细胞活力,透射电子显微镜观察线粒体形态改变,免疫组织化学法检测细胞内DRP1和OPA1蛋白表达。结果:H/R组与对照组相比,细胞活力下降,线粒体出现凋亡相关改变,OPA1表达减少而DRP1的表达增加。结论:缺氧-复氧可导致HK-2细胞发生凋亡相关改变,其机制可能与诱导线粒体形态相关蛋白DRP1、OPA1的表达改变相关。 Objective: To explore the changes of expressions of dynamin related protein 1 ( DRP1 ) and optic atrophy 1 ( OPA1 ) in epithelial cells of human proximal convoluted tubule after hypoxia/reoxygenation ( H/R ) . Methods : The epithelial cell trains of human proximal convoluted tubule were selected as study objects, then the cells were randomly divided into normal control group and H/R group. The cells in normal control group were cultured conventionally, while the cells in H/R group were cultured in anaerobic condition for 24 hours, then were cultured in aerobic condition for 6 hours. Light microscope was used to observe the change of cellular morphology, CCK - 8 was used to detect cellular activity, transmission electron microscope was used to observe the change of mitoehondrial morphology, and immunohistochemieal method was used to detect the expressions of DRP1 and OPAl in the cells. Results: Compared with control group, the cellular activity in H/R group decreased, apoptosis - related changes were observed in mitoehondria, the expression of OPAl decreased, and the expression of DRP1 increased. Conclusion: H/P can induce apoptosis - related changes of HK - 2 cells, the mechanism may be related to the changes of expressions of DRP1 and OPAl induced by H/P.
