血脑屏障模型及中枢神经系统药物转运的评价 被引量:6

摘要 目的阐述细胞模型的合理构建和中枢神经系统药物的正确评价在中枢神经系统药物转运系统开发中的重要意义。方法查阅大量近期国内外相关文献。结果与结论中枢神经系统(CNS)与血液之间存在着复杂的动态界面,即血脑屏障(BBB)。血脑屏障严格控制着血液与脑室间的物质交换,虽有利于维持脑内环境稳定,但对中枢神经系统药物转运构成了巨大的挑战。为评估药物穿透血脑屏障的能力,出现了各种体外血脑屏障模型,如细胞共培养系统和动态模型;通过血脑屏障模型评估中枢神经系统药物效应,应综合考虑游离药物浓度、药物穿透速度和程度等因素。
