通肺胶囊对烟熏加脂多糖诱发大鼠慢性阻塞性肺疾病模型的影响

Effect of Tongfei Capsule on Rat Model of Chronic Obstructive Pulmonary Disease Induced by Smoke Plus Lipopolysaccharide

【目的】观察通肺胶囊对烟熏加脂多糖诱发大鼠慢性阻塞性肺疾病模型(COPD)的影响。【方法】复制烟熏加脂多糖(lipopolysaccharide,LPS)诱发大鼠COPD模型,随机分成空白对照组,模型组,阳性对照组(盐酸班布特罗片,剂量为3.3 mg/kg),通肺胶囊低、中、高剂量组(剂量分别为1.31、2.62、5.24 g/kg);对各组大鼠进行静态肺顺应性的测定(吸气),检测肺组织匀浆丙二醛(MDA)含量、超氧化物歧化酶(SOD)活性,进行右肺及支气管细胞学计数及分类检查,测定左肺体积及进行左肺组织病理学检查。【结果】通肺胶囊各剂量组可不同程度提高模型大鼠的静态肺顺应性和肺组织SOD活力,降低肺体积比和白细胞数量,不同程度改善模型大鼠肺泡扩张、肺泡间隔断裂及肺泡融合等病理变化。【结论】通肺胶囊对烟熏加脂多糖诱发慢性阻塞性肺疾病模型大鼠具有一定的治疗作用。

Objective To observe the effect of Tongfei Capsule（TC）on rat model of chronic obstructive pulmonary disease（COPD） induced by smoke plus lipopolysaccharide（LPS）.Methods The rat model of COPD was induced by smoke plus LPS.Rats were randomly divided into blank group,model group,positive control group（bambuterol hydrochloride tablets,3.3mg/kg）,and low-,moderate-and high-dosage TC groups（in the dosages of 1.31,2.62,and 5.24 g/kg）.Static lung compliance（in air-intake state） was detected,the content of malondialdehyde（MDA） and the activity of superoxide dismutase（SOD） in lung tissue homogenate were examined.In addition,we recorded the result of the cytological count and classification examination of right lung and bronchia.Finally,we determined the volume of left lung and perform histopathological examination of left lung.Results Static lung compliance and pulmonary SOD activity were enhanced to various degrees,lung volume ratio and pulmonary white blood cell count were decreased,and alveolar expansion,alveolar septum rupture and alveolar fusion were improved to some degrees in TC groups（P0.05 or P0.01 compared with those in the model group）.Conclusion TC has certain therapeutic effect on rat model of chronic obstructive pulmonary disease induced by smoke plus LPS.
