摘要
目的:建立高效液相色谱-蒸发光散射检测器法测定妥布霉素的有关物质。方法:采用Phenomenex-Gemi-ni C18(250 mm×4.6 mm,5μm)色谱柱,以0.2 mol/L三氟醋酸(TFA)为流动相,流速为0.4 ml/min,柱温为30℃,漂移管温度110℃,载气流量为3.0 L/min。结果:有关物质卡那霉素B与妥布霉素保留时间最为接近,二者能完全分离,分离度为1.61。结论:方法专属性好,可用于妥布霉素有关物质的测定。
Objective:Todetermine related substances in tobramycin by HPLC-ELSD.Methods:The determination was performed on a C18(250 mm×4.6 mm,5 μm)column,Phenomenex-Gemini was applied as stationary phase and 0.2 mol/L Trifluoroacetic as the flow.The flow rate was 0.4 ml/min and the column temperature was 30 ℃.The drift tube temperature was 110 ℃ and the flow rate of nebulizing gas was 3.0 L/min.Results:Tobramycin and its related substance were completely separated and the resolution was 1.6.Conclusion:The method is proved to be specific and can be used for the determination of tobramycin and its related substances.
