摘要
目的分析血管紧张素Ⅱ(AngⅡ)含量、HOMA-IR胰岛素抵抗指数及血脂变化,探求持续高脂饮食及附子灌胃对自发性高血压大鼠(spontaneously hypertensive rat,SHR)内皮功能及糖脂代谢的影响,并探讨活血潜阳方的干预作用。方法以5周龄雄性SHR为对象,通过持续高脂饮食及附子灌胃4周,随机分为治疗组及模型对照组。并以年龄和性别相匹配的空白SHR大鼠及WKY大鼠对照。治疗组以活血潜阳方灌胃。其余各组以纯净饮用水灌胃。治疗8周后腹主动脉取血,测定AngⅡ含量,空腹血糖、胰岛素、三酰甘油(TG)及胆固醇(TC)水平。结果各组SHR大鼠的AngⅡ含量、HOMA-IR指数、TG及TC均明显高于WKY组;其中高脂饮食+附子灌胃的SHR大鼠模型组(SHR-C)各指标进一步显著提高,而活血潜阳方治疗8周,可以显著改善上述指标。结论高脂饮食+附子灌胃可以造成SHR大鼠更严重的内皮功能损伤,血脂异常及胰岛素抵抗。活血潜阳方可有效降低AngⅡ含量,改善其血脂异常及胰岛素抵抗状态。
Objective To explore the effect of Huoxue Qianyang formula(HQF) on the endothelial function and glycolipid metabolism in spontaneously hypertensive rats(SHRs).Methods The 5 weeks old male SHRs fed with monkshood for 4 weeks and high-fat diet constantly.They were randomly divided into two groups:Untreated group and HQF treated group.The sex and age matched Wistar-Kyoto rats(WKY) and another 11 SHRs served as controls.The drugs were administered by intragastric administration two times daily in the HQF treated group.After 8 weeks,the angiotensin Ⅱ(AngⅡ),fasting blood-glucose,insulin,cholesterol and triglyceride were examined.Results The levels of serum AngⅡ,homeostasis model assessment of insulin resistance(HOMA-IR),cholesterol and triglyceride were significantly higher than that in WKY rats.The levels of HOMA-IR,cholesterol and triglyceride in SHRs fed with high-fat diet and monkshood,were significantly higher than that in others.After treatment with HQF,the levels were significantly lowered.Conclusion High-fat diet and intragastric administration of monkshood caused the SHRs with more severely impaired endothelial function,dyslipidemia and insulin resistance.HQF could reduce the level of serum AngⅡ and improve the glycolipid metabolism.
