Rho激酶抑制剂对创伤性脑损伤后脑细胞内亚二倍体比率的影响及其意义

摘要 目的探讨颅脑损伤后应用Rho激酶抑制剂对大鼠脑创伤区皮层细胞亚二倍体比率的影响及意义。方法健康成年雄性Wistar大鼠45只,随机分为3组,即假手术组(Sham组)、创伤组(TBI组)和Rho激酶抑制剂干预组(FSD组)。Feeney法制作自由落体创伤性脑损伤(TBI)动物模型,造模打击后,即刻给予FSD组大鼠尾静脉注射Rho激酶抑制剂法舒地尔(1mg/kg)2mL,Sham组和TBI组给予注射用水2mL做对照,伤后6h处死大鼠,取脑。采用流式细胞仪(FCM)碘化丙啶单染色法检测鼠脑创伤区皮层细胞亚二倍体比率。结果 Sham组、TBI组和FSD组的创伤区皮层细胞亚二倍体比率分别为(1.58±0.35)%,(15.90±3.91)%和(5.35±2.10)%,3组间差异有统计学意义(P<0.01)。结论 Rho激酶抑制剂对TBI后大鼠创伤区脑组织细胞亚二倍体比率大小存在影响,能够抑制TBI后损伤区脑细胞的早期凋亡,减轻创伤后的脑神经功能障碍。
