结直肠癌内镜活检组织p53基因变异的PCR-SSCP分析

The Mutations of p53 Gene Detected by PCR-SSCP in Biopsy Samples of Colorectal Carcinomas

目的 探讨结直肠癌p5 3基因变异情况及其意义。方法 采用PCR技术扩增内镜下活检的结直肠癌组织中p5 3基因第 5～ 9外显子 ,再以银染色单链构象多态性 (SSCP)技术分析其变异情况。结果 2 0例结直肠癌组织 p5 3基因的PCR扩增均获成功 ;银染色SSCP分析发现其中 5例有异常条带 ,阳性率为 2 5 .0 % ,主要见于第 5、7外显子 ,第 9外显子未见阳性带 ;3例升结肠癌中有 2例阳性。结论 结直肠癌 p5 3基因变异率不高 ,但研究 p5 3基因变异有助于认识结直肠癌的发生发展规律。

Objective To investigate the p53 gene mutations in colorectal carcinomas and its significance.Methods The exon 5～9 of p53 gene in endoscopic biopsy tissues of colorectal cancer were amplified by polymerase chain reaction(PCR),and its mutations were detected by single strand conformation polymorphism(SSCP) with silver staining.Results p53 gene was successfully amplified by PCR in all 20 cases,and five of them(25.0%) were positive in SSCP analysis.Positive results were mainly found in exon 5 and exon 7,none in exon 9.Two of three ascending colon cancer were positive.Conclusion Our results show that the mutation rate of p53 gene is low in colorectal carcinoma,but study on p53 gene mutation is valuable for realizing the development of colorectal carcinoma.
