探讨miRNA-193b和miRNA-100在乳腺癌中的差异表达 被引量:1

摘要 目的采用已知的miRNA(microRNA)基因中选定的目标基因,筛选与乳腺癌相关miR-NAs,并采用real time-PCR方法验证基因,探讨其在乳腺癌发生发展中的作用,研究结果为探明miR-NA在乳腺癌中的差异表达及乳腺癌的基因诊断和治疗提供实验依据。方法收集乳腺癌组织及对照的癌周组织10对,分别提取RNA。对乳腺癌中的差异表达miRNA基因进行验证。从中选取表达上调明显的miRNA-193b及下调明显的miRNA-100,采用实时定量PCR技术进一步验证结果。结果证实miRNA的real time-PCR结果与已知基因结果基本吻合,miR-193b是乳腺癌中上调的miRNA基因,miR-100是乳腺癌中明显下调的miRNA基因。结论对miRNA基因与乳腺癌的关系有进一步深入了解,相对于癌周组织进一步验证了1个表达上调和1个表达下调的miRNA基因,表达上调基因可能为潜在的致癌基因,表达下调基因可能为潜在的抑癌基因。
