摘要
目的 研究腺病毒早期转录单位增强子结合蛋白 (E1AF)对膀胱癌细胞侵袭能力的影响。 方法 构建真核表达载体pcDNA3 E1AF ,并以此载体转染膀胱癌细胞株BIU 87。采用Boy denChamber穿膜实验分别检测未转染的BIU 87细胞、转染后细胞和转染空载体细胞的体外侵袭能力。将未转染的BIU 87细胞和转染后的细胞分别接种至裸鼠进行体内实验。 结果 转染后细胞的侵袭能力较转染前明显增强 ,肿瘤细胞图像分析和PCNA的表达 ,均显示转染后的肿瘤细胞增殖明显活跃。 结论 E1AF可以显著增强膀胱癌细胞的侵袭能力及增殖能力 。
Objective To investigate the influence of alien expressive E1AF on invasiveness of the bladder carcinoma cells. Methods An eukaryotic expressive vector pcDNA3 E1AF was constructed and introduced into a strain of bladder carcinoma cells BIU 87 with transfection. The invasiveness in vitro of the transfected cells was studied by means of the Boyden Chamber test and tumorigenicity in nude mice was investigated. Meanwhile, transfected group with empty vector and untransfected group were set up as control. Results The cells transfected with E1AF is remarkably more invasive than those of control group. The image analysis and PCNA expression of the tumor cells demonstrate that transfected cells are more actively proliferative than the untransfected cells. Conclusions E1AF enhances remarkably the invasiveness in vitro and the proliferativeness of the bladder carcinoma cells and might play an important role in the invasiveness and metastasis of the bladder carcinoma cells.
