The significance of bFGF mRNA expression in benign prostatic hyperplasia

摘要 目的 探讨前列腺组织中碱性成纤维细胞生长因子 (bFGF)基因和蛋白及其受体表达的意义。 方法 应用原位分子杂交和免疫组化方法检测 38例前列腺增生 (BPH)组织和 1 0例正常前列腺 (NP)组织中bFGFmRNA、bFGF和FGFR1表达。 结果 前列腺间质细胞和上皮细胞中均见bFGFmRNA、bFGF和FGFR1阳性染色。BPH间质细胞中三者表达显著高于NP组织 ,但BPH与NP上皮细胞中三者表达差异无显著性。BPH间质细胞中bFGFmRNA与bFGF共同表达率达 82 % ,而上皮细胞中仅为 42 % ;BPH间质细胞中bFGF与FGFR1共同表达率为 71 % ,上皮细胞中为 45%。 结论 bFGF FGFR通道在BPH发生尤其是间质细胞增生过程中起着重要作用 ; Objective To investigate the distribution and expression of bFGF mRNA, bFGF and FGFR1 in normal and hyperplastic prostates. Methods In 38 specimens of benign prostatic hyperplasia(BPH) and in 10 normal prostates tissues (NP),bFGF mRNA expressions was examined by in situ hybridization, bFGF and FGFR1 by immunohistochemistry. Results Positive stain of bFGF mRNA, bFGF and FGFR1 were observed in both stroma and epithelium of the examined prostates.The expressions in stroma of BPH were significant higher than these in stroma of NP.Whereas the expressions in epithe lium revealed no significant difference bewteen BPH and NP.Coexpression rate of bFGF mRNA and bFGF in stroma of BPH was 82%,whereas only 42% in the epithelium of BPH.Coexpression of bFGF and FGFR1 was 71% in stroma and 45% in epithelium of BPH. Conclusions bFGF FGFR pathway plays an important role in the pathogenesis of BPH,expecially in stroma hyperplasia.Abnormal expression of bFGF in stroma of BPH is mainly resulted from the increase of gene transcription.
