不同环氧合酶-2抑制剂治疗大鼠脑外伤早期炎症实验观察 被引量:3

摘要 目的 :探讨环氧合酶-2(COX-2)抑制剂尼美舒利(Nimesulide)和吲哚美辛(Indom-ethacin)在减轻脑外伤早期炎症反应中的治疗作用。方法:采用液压冲击法建立实验动物脑创伤模型,每间隔12h腹腔注射尼美舒利和吲哚美辛,分别于伤后24、48、72和96h应用RT-PCR和Western-Blot法检测环氧合酶-2mRNA和蛋白水平,ELISA法检测前列腺素E2(PGE2)水平,实验动物给予以神经功能评分。结果:选择性COX-2抑制剂尼美舒利在降低脑外伤早期环氧化酶-2mRNA及蛋白质合成方面强于非选择性COX-2抑制剂吲哚美辛,但二者在降低PGE2水平以及改善神经功能方面无显著性差异(P>0.05)。结论:COX-2抑制剂尼美舒利和吲哚美辛在脑外伤早期治疗中的抗炎作用相当,但吲哚美辛的抗炎机制可能涉及到更多的炎症通路。 Objective:To compare anti-inflammatory effect between Nimesulide and Indomethacin in early traumatic brain of rats.Methods: To establish animal models of traumatic brain injury in rats,and intraperitoneal injection with nimesulide and indomethacin respectively per 12 hour.At the time point of 24,48,72 and 96 hour after the injury,the brain tissue was obtained for the using of RT-PCR and Western-Blot,and detected mRNA and protein level expression of cydooxygeaase-2(COX-2),then prostaglandin-E2(PGE2) level was detected by enzyme linked immunosorbent assay(ELISA),and all the rats were given a mark for their nerve function.Results: The results from RT-PCR and Western-Blot in dicated that the expression of COX-2 in mRNA and protein level in nimesulide-treated group was lower than those in domethacin-treated group,but the PGE2 level and nerve function score between them had no statistical significance.Conclusion:The two COX-2 inhibitors,nimesulide and indomethacin,have equal anti-inflammatory effect in early traumatic brain tissue,and the latter may be involve more inflammation passageway.
