摘要
研究超声波辅助碱提酸析法提取禾雀花黑色素工艺。以黑色素标准品A400 nm吸光度法检测禾雀花黑色素含量,单因素试验筛选碱提酸析法和L(943)正交试验筛选超声波辅助碱提酸析法提取禾雀花黑色素最佳工艺。黑色素标准品A400 nm吸光度法检测禾雀花黑色素含量简单、准确;禾雀花黑色素含量(6.89±1.7)%;碱提酸析法最佳工艺为1.0 mol/L NaOH溶液、料液比1∶15(g/mL),浸提3 h 2次,粗提物得率(93.3±1.8)%,黑色素含量(34.7±1.5)%;超声波辅助碱提酸析法最佳工艺为料液比1∶15(g/mL)、1.0 mol/L NaOH溶液、300 W超声波处理40 min,粗提物得率(94.8±1.5)%,黑色素纯度(35.4±1.2)%。禾雀花含有丰富的植物黑色素,用UA-AEAP法提取禾雀花黑色素工艺简单易行,值得进一步研究和开发利用。
Research methods of extracting melanin from flowers of Mucuna Birdwoodiana Tutch.The melanin content was analyzed with the reference of standard melanin by spectrophotometry to detect A400 nm;the optimum conditions of alkaline-extraction and acid-precipitation method were screened by the single factor trial,and the conditions of the ultrasonic-assisted alkaline-extraction and acid-precipitation method were optimized by the orthogonal test of L9(43).Results: The spectrophotometry to analyze the melanin content was simple and workable;the melanin content of the flowers of Mucuna Birdwoodiana Tutch.was(6.89±1.7) %;The optimal conditions of alkaline-extraction and acid-precipitation method were: 1.0 mol/L NaOH solution,material : solution =1∶15(g/mL),extracting 3 h for 2 times,the recovery rate of the crude product was(93.3±1.8) %,and the melanin content was(34.7±1.5) %;The optimal conditions of ultrasonic-assisted alkaline-extraction and acidprecipitation method were: 1.0 mol/L NaOH solution,material : solution =1∶15(g/mL),300 W ultrasonic treating 40 min,the recovery rate of the crude product was(94.8±1.5) %,and the melanin content was(35.4±1.2) %.The flowers of Mucuna Birdwoodiana Tutch.contented rich melanin and deserved further research and development,and the extraction time and the technology of extraction of melanin from the flowers of Mucuna Birdwoodiana Tutch.with alkaline-extraction and acid-precipitation method shortened and simplified remarkably by the ultrasonic-assisted treatment.
