摘要
目的:探讨滋补肝肾方药"复健片"对大脑中动脉闭塞(MCAO)模型大鼠皮质脊髓束重塑作用。方法:Wistar大鼠随机分为假手术组、模型组和药物组,各组根据观测时间点再分为术后4周末、术后5周末两个亚组。采用电凝法闭塞大鼠右侧大脑中动脉,药物组术后1周末按9 g生药/kg给予复健片水溶液灌胃,于术后3周末注射BDA,采用走横木实验(BWT)对各组大鼠运动功能进行评分;采用免疫组织化学方法观测大鼠左侧颈髓BDA和GAP-43的表达。结果 :模型组和药物组BWT评分随着术后时间的延长而升高(P<0.01),其中药物组的评分升高更显著,至术后5周末,与假手术组已无明显差异(P>0.05);而术后4周末,药物组和模型组的左侧颈髓GAP-43平均灰度值较假手术组显著降低(分别为P<0.05、P<0.01),至术后5周末,模型组GAP-43平均灰度值较之同期假手术组,已无显著差异(P>0.05),药物组GAP-43平均灰度值则明显低于其余两组(P<0.01);与同期假手术组相比,模型组和药物组左侧颈髓BDA平均灰度值均明显降低(P<0.01),其中药物组BDA平均灰度值降低更为明显(与同期模型组相比,P<0.05)。结论 :复健片既可以延长MCAO大鼠皮质脊髓束的重塑时间,又可以促进更多的皮质脊髓纤维束越过中线,支配同侧颈髓运动神经元。这可能是复健片促进缺血性脑卒中后期运动神经功能恢复的又一重要机制。
Objective:To investigate the effects of Fujian tablet on corticospinal tract remodeling of middle cerebral artery occlusion(MCAO)rats.Methods:Wistar rats were randomly divided into three groups:sham surgery group,model group and administration group.Each group was randomized into two subgroups according to operation time at the end of 4th week or 5th week.The right middle cerebral artery of models were occluded by electrocoagulation.Fujian tablet docoction was intragastrically perfused at a dose of 9 g crude drug/kg at the end of 1 week after surgery in administration group,BDA was injected into the left frontal motor cortex at the end of 3th weeks after surgery.Beam walking test(BWT) was performed to estimate the motor function.Immunohistochemistry was used to measure the expressions of BDA and growth-associated protein(GAP-43) in the left cervical spinal cord.Results:BWT score of model group and administration group increased with the extension of time(P0.01).Administration group score increased more significantly,at the end of 5th week,there was no difference between sham operation and administration group(P0.05).At the end of 4th week,compared with sham operation group,mean grey values of the expression of GAP-43 reduced in the left cervical spinal cord of model group(P0.05) and administration group(P0.01).At the end of 5th week,mean grey values of the expression of GAP-43 of model group had no difference from sham operation group(P0.05),while those of administration group were lower significantly than other two groups(P0.01).Meanwhile,compared with sham operation group,mean grey values of the expression of BDA in other two groups reduced remarkably(P0.01),especially in administration group(P0.05),vs model group at the same time.Conclusion:Fujian tablet can not only prolong the corticospinal tract remodeling of MCAO rats,but also promote more corticospinal fibers originating from the intact cerebral cortex crossed the midline to enhance the innervation of stroke-impaired peripheral tissues,which maybe an important mechanism of Fujian tablet to promote motor function recovery following cerebral ischemia.
