摘要 目的:研究慢性根尖周炎感染根管内中间普氏菌是否存在基因多态性。方法:选择13例慢性根尖周炎患者,分别取其感染根管内样本,分离培养,经中间普氏菌特异引物PCR鉴定的克隆,再利用随机引物PCR(AP-PCR)检测其基因多态性。结果:13例慢性根尖周炎样本中3例分离出中间普氏菌,共95株,并检测出3种基因型,其中A、B、C样本各检出一种基因型。结论:慢性根尖周炎患者感染根管内中间普氏菌存在基因多态性。 Objective: To study the clonal diversity of Prevotella intermedia strains isolated from infected root canals of teeth with chronic periapcial periodontitis(CAP).Methods: Samples from the infected root canals of the teeth with CAP were obtained in 13 patients by sterilized paper points.The samples were cultured using selective medium including CDC anaerobic blood agar base and wancomycin and kanamycin.Black-pigmented clones were selected,DNA of the clones were extracted and identified by using a P.intermedia species-specific 16S rRNA genebased polymerase chain reaction assay.Recovered P.intermedia isolates were analyzed for clone diversity by using arbitrarily primer PCR.Results: Black-pigmented clones were isolated from 3 infected root canals out off the 13 samples.95 clones were collected and identified by using P.intermedia species-specific PCR,including 32 clones from A sample,36 clones from B sample and 27 clones from C sample.Typing of P.intermedia colonies retrieved from 3 infected root canals revealed 3 genotypes.One genotype was detected from each of A,B and C sample respectively.Conclusion: Gene polymorphism of P.intermedia presentes in root canals of the teeth with CAP.
