摘要
目的:建立人血浆中麦考酚酸(MPA)含量的高效液相色谱法(HPLC法),并用该方法对肾移植患者血浆中的浓度进行测定。方法:Diamonsil C18色谱柱(250 mm×4.6 mm,5μm);流动相:乙腈-0.1%三氟乙酸(40∶60),流速为1.0 mL.min-1,柱温为35℃,紫外检测波长为215 nm,血样用乙腈沉淀蛋白法进行处理。结果:麦考酚酸浓度在0.20~49.96μg.mL-1(r=0.999 6)范围内线性关系良好,最小检测浓度为0.20μg.mL-1,平均回收率均>90%,提取回收率>100%,批间和批内RSD<15%。结论:本方法准确、简便、重现性好,可用于麦考酚酸的血药浓度监测及药动学研究。
OBJECTIVE:To establish a high-performance liquid chromatography(HPLC) method for determination of plasma concentration of mycophenolic acid in patients undergoing renal transplantation.METHODS:Mycophenolic acid was determined using Diamonsil C18 column(250 mm×4.6 mm,5 μm) with mobile phase consisting of acetonitrile-0.1% trifluoroacetic acid(40∶60) at a flow rate of 1.0 mL·min-1.The column temperature was maintained at 35 ℃ and the UV detection wavelength was set at 215 nm.Plasma samples were treated by precipitation of protein using acetonitrile.RESULTS:The linear concentration of mycophenolic acid was 0.2-50.0 μg·mL-1(r=0.999 6) with minimum detectable concentration of 0.2 μg·mL-1.The average recoveries rates were all greater than 90%,with extraction recovery rates greater than 100%,and relative standard deviation(RSD) of interassay and intrassay all less than 15%.CONCLUSION:The method is proved to be accurate,simple,and recurrent and it is suitable for the blood concentration monitoring and pharmacokinetic study of mycophenolic acid.
