摘要
目的探讨紫外线照射对果蝇凋亡相关基因reaper(rpr),grim,hid表达的影响。方法收集8 h内羽化果蝇,雌雄分开培养至10日龄,紫外线照射0,0.5,1,1.5,2 h。提取果蝇总RNA,运用逆转录PCR(RT-PCR)方法检测各组果蝇rpr、grim、hid基因的表达。结果对照组果蝇rpr、grim、hid基因表达较弱,照射后rpr、grim、hid基因的表达量呈现先增后减的趋势。结论紫外线照射可增加果蝇凋亡相关基因rpr,grim,hid的表达,可能是其诱导细胞凋亡的分子机制之一,但照射达到一定时间,基因表达减弱,可能是其引起衰老的原因。
Objective To study the effect on the expressions of apoptosis related genes reaper( rpr), grim, hid in Drosophila mela- nogaster after ultraviolet irradiation. Methods The flies of Drosophila were collected within 8 hours after eclosion. After cultured on standard medium for 10 days, flies were exposed to ultraviolet for different times (0. 0 ,0. 5,1.0,1.5,2. 0 h). Then the total RNA from flies were extracted. The expressions of genes rpr, grim, hid were detected by Reverse Transcription PCR (RT-PCR). Results In the control group, the expressions of apoptosis related genes rpr, grim, hid in Drosophila were weak. After ultraviolet irradiation, the expressions of genes rpr, grim, hid were increased at first and then decreased. Conclusions Ultraviolet irradiation can enhance the expressions of apoptosis related genes rpr, grim, hid in Drosophila, which may be one of the molecular mechanisms of apoptosis induced by ultraviolet. But after a certain time expo- sure, expressions are decreased, which may be the cause of aging.
