摘要
为了提高木瓜蛋白酶的稳定性,以壳聚糖为载体,研究了壳聚糖纯化条件及木瓜蛋白酶固定化条件。最终确定采用0.1%的乙酸和5mol/L的NaOH对壳聚糖纯化,并用冷冻真空干燥的方法对壳聚糖进行干燥。确定最佳固定化条件为:交联剂浓度为0.4%,pH值为7.0,加酶量为45mg/g壳聚糖,固定时间为12h。经固定化后,酶稳定性能有所提高。
The purpose of the study was to promote papain enzyme stability. The ehitosan was used as carrier in this research. The purification condition of chitosan and the immobilization method of papain enzyme were studied. The results showed that 0.1% acetic acid and 5mol/L NaOH could be used to purify chitosan. The pure chitosan should be dried under cold vacuum drying condition. The proper immobilization condition was as follows: formaldehyde concentration was 0.4%, pH was 7.0, enzyme content was 45mg/g chitosan, and immobilization time was 12h. After immobilization, the enzyme stability was enhanced.
