摘要
目的 :为了得到新的抗HSV中药方剂的最佳配方比 ,我们进行了此项研究。方法 :我们以阿昔洛韦 (ACV)为阳性对照 ,采用空斑减数实验 ,观察了虎杖、黄芪单用及合用抗HSV- 2 333株的药效 ,用中效作用原理对药效进行了分析。结果 :在HEp 2 细胞系统中 ,ACV对HSV- 2 333株直接杀灭、增殖抑制、感染阻断的ED50 依次为 16 .4 5、10 .85、18.6 2 μg/ml,虎杖黄芪合用比为 1(Dm )虎杖 / 1(Dm )黄芪时 ,相应数值分别为 2 .2 7、2 .0 4、1.32 μg/ml。与ACV相比 ,药效比ACV强 ,而且合用指数 (CI)平均值最小 ,协同作用最明显。结论 :上述合用比对HSV- 2 333株直接杀灭、增殖抑制、感染阻断的治疗指数 (TI )是ACV的 72 6、52 5、140 0倍 ,是组方时的最佳配比。
Objective:In order to obtain the most excellent dispensing proportion of Chinese medicinal herbs,we carry out this research.Method:we use ACV as positive control,observe antiherpes effects on HSV -2 333 strain of rhizoma polygoni cuspidati and astragalus membranaceus which are used lonely or in combination byplaque reduction assay,and make effect analysis of combination by median effect principle.Result:In HEp -2 cell system,the ED 50 s of the direct virucidal activity,the inhibition effects on the multiplication and infection of HSV -2 333 strain of ACV are 16.45,10.85 and 18.12μg/ml respectively.When combination ratio of rhizoma polygoni cuspidati/astragalus membranaceus equals 1(ED 50 ) of the former/1(ED 50 ) of the latter,the corresponding figures are 2.27,2.04 and 1.32μg/ml separately.Comparing with ACV,the effects is stronger than ACV,and the average combination index is the smallest,there is the most significant synergism.Conclusion:The treatment indices(TI) of the direct virucidal activity,the inhibition effects on the multiplication and infection of HSV -2 333 strain of this combination ratio are 726,525,1 400 times of ACV TIs respectively,it is the most excellent combination ratio.
