摘要
目的 探讨周围神经损伤后脊髓运动神经元表达睫状神经营养因子 (CNTF)水平的变化规律及其与神经元病理变化的相关性。 方法 选用出生后 3d和成年大鼠各 30只 ,均分为对照组、伤后 3,7,14,2 1,2 8d 6组。切断不同鼠龄Wistar大鼠坐骨神经复制周围神经损伤模型 ,行免疫组织化学染色CNTF阳性脊髓运动神经元 ,利用图像分析系统计算脊髓CNTF阳性运动神经元的吸光度值 ,从而间接反映脊髓运动神经元CNTF水平。 结果 坐骨神经损伤后腰段脊髓运动神经元CNTF水平很快下降 ,至伤后第 2周降至最低水平 ,随后逐渐恢复 ,至 4周接近对照组水平 ;光镜病理改变情况与此过程相似。 结论 周围神经损伤后 ,脊髓运动神经元表达CNTF水平将发生变化 ,而且其变化规律与损伤后运动神经元的病理变化过程相似 ;运动神经元内源性CNTF水平与其病变程度呈负相关。
Objective To study the change of ciliary neurotrophic factors (CNTF) in spinal motor neurons and its relationship with the degree of degeneration and death of the spinal motor neurons following sciatic nerve injury in rats. Methods The sciatic nerve injury was induced by cutting and removing part of the sciatic nerve in the adult Wistar rats aging 3 d. The rats were allowed to survive for 3, 7, 14, 21 and 28 d, respectively. The extent of the death and degeneration of the spinal motor neurons was examined, and the level of CNTF in the spinal motor neurons was also measured with image analysis. Results The level of CNTF in the spinal motor neurons was reduced following sciatic nerve injury, and it was lowest in the 2nd week after injury. Then it increased gradually, and reached the normal level on the 28th d following sciatic nerve injury. The change of the spinal motor neurons under light microscopy was similar to that of CNTF. Conclusions The level of CNTF in the spinal motor neurons changes following sciatic nerve injury, and the death and degeneration of the spinal motor neurons may be induced by the peripheral nerve injury. The level of CNTF in the spinal motor neurons is negative with the extent of the death and degeneration of the spinal motor neurons.
