摘要
目的对壳二孢氯素产生菌F05Z0761进行菌种鉴定,并进行培养基优化,提高其发酵单位。方法首先通过形态学特征进行鉴定,然后通过发酵培养基筛选实验并采用Plackett-Burman实验、最陡爬坡实验、中心组合实验设计等方法提高发酵单位。结果 F05Z0761经形态学鉴定显示该菌株属于镰刀菌属(Fusarium),并得到了优化后的发酵培养基配方即:淀粉1.8%、葡萄糖2.5%、棉籽粉1.9%、热榨黄豆饼粉0.8%和KH_2PO_40.2%,发酵单位较对照提高了4.38倍。结论由镰刀菌属产生壳二孢氯素在国内还未见报道,并将响应面实验设计应用于该菌株的发酵培养基优化中。
Objective To identify the filamentous fungus F05Z0761 which is able to produce ascochlorin and to improve the fermentation titer. Methods Morphological observation was applied to identify the strain and Plackett-Burman design, steepest ascent design, central composite design were used to improve the fermentation titer. Results The strain was identified as Fusarium by morphological characteristics and the optimum medium composition for ascochlorin production was found to be: Starch 1.8%; Glucose 2.5%; Pharmamedia 1.9%; Soybean meal 0.8% and KHEPO4 0.2%. The fermentation titer was improved 4.38 times as much as the initial titer. Conclusion This is the first report that ascochlorin could be produced by Fusaium in China. And response surface methodology was used to optimize the fermentation medium
