高效毛细管电泳法测定加味逍遥丸中芍药苷、栀子苷的含量 被引量:9

Determination of Paeoniflorin and Geniposide in Jiawei Xiaoyao Pills by High Performance Capillary Electrophoresis

摘要 目的:建立加味逍遥丸中芍药苷、栀子苷同时测定的高效毛细管电泳分析方法。方法:采用高效毛细管电泳法,缓冲体系为25 mmol·L-1硼砂缓冲液(NaOH调pH 10.5),熔融石英毛细管(60 cm×50μm),运行电压25 kV,柱温25℃,检测波长230 nm。结果:芍药苷、栀子苷分别在0.008 1~0.162 6 g·L-1和0.023 8~0.475 6 g·L-1线性关系良好(r=0.999 8和0.999 7),样品平均加样回收率为99.67%(RSD 1.27%)和99.36%(RSD 1.53%)结论:该方法简单、快速准确、重复性好,适用于加味逍遥丸的质量控制。 Objective: To establish a new high performance capillary electrophoresis (HPCE) method for determination of paeoniflorin and geniposide in Jiawei Xiaoyao pills. Method: Paeoniflorin and geniposide in Jiawei Xiaoyao pill were determined by using HPCE. The background electrolyte system selected was 25 mmol. L- sodium borate buffer (pH was adjusted to 10.5 by caustic soda). Capillary electrophoresis was performed using 60 cm ×50 μm fused silica capillary column, applied voltage was 25 kV; temperature was 25 ℃. The detection wavelength was set at 230 nm. Result: The linear range of paeoniflorin and geniposide was 0. 008 1-0. 162 6 g · L^-1 and 0.023 8-0.475 6 g · L^-1 (r =0.999 8 and 0.999 7), and the average recovery was 99.67% (RSD 1.27% ) and 99.36% (RSD 1.53% ). Conclusion: The method is simple, rapid and reproducible, and could be used for the quality control of Jiawei Xiaoyao pills.
