摘要
以"大团蜜露"水蜜桃果实为试材,通过酶联比色法对水蜜桃果实中磷脂酶D活性进行了测定,并对影响酶活性测定结果的因素进行了研究,建立了用于测定桃果实组织磷脂酶D活性的方法。在200μL反应体系中,膜蛋白含量和浓度分别以10μg和10mmol/L为宜。通过分析正丁醇和水杨酸处理的水蜜桃在低温贮藏中果实样品酶活测定结果得出,该测定方法具有高效便捷、灵敏度高、危害低和结果稳定的优点,可用于桃果实组织中磷脂酶D活性变化的研究。
An improved method for detecting phospholipase D(PLD) activity was proposed by using the enzyme coupling colorimetric method to experiment on the honey peaches("Datuanmilu" of Shanghai Nanhui district).The influencing factors of detecting PLD activity were studied.In 200 μL reaction system,the proper protein content and Ca2+ concentration is 10 μg and 10 mmol/L.The effects of post-harvest treatment with n-butyl alcohol and salicylic acid of PLD activity of honey peaches were studied under 4 ℃ storage.The results showed that the enzyme coupling method is simple,accurate,low-hurt and stabilize,and it can be used in the determination of activity changes of PLD in tissues of peach.
