大孔吸附树脂分离纯化甘草中α-葡萄糖苷酶抑制剂的研究 被引量:4

摘要 目的:筛选纯化甘草α-葡萄糖苷酶抑制物质的最佳大孔吸附树脂,并对影响纯化的各种因素进行系统的研究,为工业化生产提供参考。方法:通过检测大孔树脂吸附甘草液中α-葡萄糖苷酶抑制物质的量,以α-葡萄糖苷酶抑制物质酶活性抑制率为指标对工艺进行评价,确定甘草中α-葡萄糖苷酶抑制物质的最佳纯化工艺。结果:OU-2型大孔树脂纯化效果最好,其最佳工艺为药材与树脂的比例为1/25、pH为5~6,吸附完全后,先以水洗脱,再以3BV70%乙醇洗脱。经OU-2处理后的甘草α-葡萄糖苷酶抑制物质酶活性抑制率高达95%。结论:此法简单可行,分离效果好,能满足大生产的要求。 Objective:To separate and purify of α-glucosidase inhibitor in Glycyrrhiza uralensis Fisch by macroporous adsorption resin.Methods:The adsorption and desorption capacities of macroporous resins were investigated through examining the contents of the α-glucosidase inhibitor and the effects of several influencial factors on the abosorption of α-glucosidase inhibitor by macroporous resin were studied.Results:Among 8 types of resins tested,OU-2 was shown to be the best.one for purification of α-glucosidase.The optimum ratio of herb to macroporous resin and pH value for OU-2 absorption were 1/25 and 5~6,respectively.After adsorption,the resin was eluted by water and 3 BV 70% ethanol.After OU-2 purification,the inhibitory activity of Glycyrrhiza uralensis Fisch against α-glucosidase reached 95%.Conclusion:This research provided references for large-scale production of α-glucosidase inhibitor of Glycyrrhiza uralensis Fisch.
