红细胞贮存时间对脂多糖诱导的中性粒细胞炎症释放的影响 被引量:1

摘要 目的研究红细胞(RBC)随保存时间的延长对受者中性粒细胞(PMN)炎症释放水平的影响。方法分别将红细胞在保存的第1、21、35天(D1、D21、D35)分离上清,-20℃冻存备用。将冻存的上清与健康受者PMN体外培养12 h后,加入脂多糖(LPS)后培养12、24 h收集上清冻存待检。用ELISA方法检测其IL-6和TNF-α水平。结果红细胞在LPS的刺激下IL-6水平随保存时间延长而增高,差异有统计学意义(P<0.05),TNF-α水平也有所增高,但差异无统计学意义(P>0.05)。结论在LPS的刺激下,D35较之D1红细胞上清促使PMN释放更多的IL-6和TNF-α,可以解释为输注保存时间长的红细胞增强了PMN的炎症反应,这对预防患者在大量输血后引发的相关免疫反应有一定作用。 Objective To study the effects of RBC storage time on PMN inflammation release level induced by LPS.Methods The supernatant was isolated from the RBC at different time points(D1,D21 and D35) respectively,and cryopreserved at-20℃.Freeze-stored supernatant was incubated with PMN from healthy donors for 12 h,then lipopolysaccharide(LPS) was added and cultured for 12 h and 24 h,then cells supernatants were collected.Level of IL-6 and TNF-α were detected by ELISA.Results The IL-6 level increased obviously along with the increase of RBC storage time by LPS stimulation,and there was statistically significant difference(P〈0.05).The TNF-α level had little change,but there was no significantly statistical difference(P〈0.05).Conclusion The D35 RBC supernatants release more IL-6 and TNF-α secreted from PMN than D1 induced by LPS stimulation.It can interpret that the inflammatory response in recipients upon LPS induction is enhanced by the RBC transfusion with the elongation of the RBC duration.It is useful for preventing kinds of immune responses after massive blood transfusions.
