摘要 目的探讨基质金属蛋白酶-13(MMP-13)、白细胞介素-l(IL-1)在去势大鼠退变腰椎间盘组织中的表达及临床意义。方法将30只3月龄Sprague-Dawley(SD)大鼠,随机均分为基础对照组(BL组)、去卵巢手术组(OVX组)、假手术组(sham组),每组各10只;BL组在手术开始前处死大鼠,其余两组术后3个月处死大鼠,并与处死前10d和4d分别给予显色双荧光标记;L2-4椎体进行骨密度及硬组织切片分析,k4-5,进行HE常规染色、VG特殊染色及免疫组化检测MMP-13、IL-1的表达,观察椎间盘的病理学改变并据评分标准对腰椎间盘的退变程度(LVD)进行评分。结果大鼠腰椎骨量的下降及椎间盘的退化,OVX组较Sham组严重(P〈0.05):免疫组化结果显示。OVX组与sham组比较,大鼠椎间盘中MMP-13、IL-1表达明显升高(P〈0.05)。结论MMP-13、IL-1表达量的增加是腰椎间盘退变形成和发展的重要因素。 Objective To investigate the expression of MMP-13 and IL-1 in degeneration lumbar intervertebral disc orga- nization in the ovariectomized (OVX) rats. Methods 30 SD rats of 3-months-old were randomized into the baseline (BL) group, OVX group and sham group, each group had 10 rats. Rats of the BL group were killed at the beginning of the ex- periment. Rats of the OVX and sham group were killed at 3 months after surgery. Coloration double labeling was performed at 10 d and 4 d before killed. Bone mineral density (BMD) measurement and bone histomorphometric analysis were applied in the I~ centrum. L4-5 lumbar spines and intervertebral disc sections were stained with HE, VG staining, and the expres- sion of MMP-13, IL-1 were detected by immunohistoehemistry. The pathological changes of intervertebral discs were ob- served and degree of lumbar intervertebral disc degeneration (LVD) was scored by a histological scoring system. Results The degression of spine bone mass and the degeneration of intervertebral disc in the OVX group were more serious than those in the sham group (P 〈 0.05). The expression of MMP-13 and IL-1 in the OVX group were higher than those in the sham group (P 〈 0.05). Conclusion The increased amount of MMP -13, IL-I expression are key factors in the develop- ment and progression of the lumbar intervertebral disc degeneration.
