摘要
研究野生型 p5 3基因对染石英尘肺泡巨噬细胞 (AM)培养上清导致的成纤维细胞非正常增殖和功能的影响。将体内、体外染尘 AM上清加入人胚肺成纤维细胞 (2 BS)中培养 2 4小时后 ,应用脂质体介导转染法将野生型 p5 3基因导入(2 BS)继续培养 2 4小时。结果 ,转染 p5 3基因组 2 BS细胞增殖和胶原合成较未转染组受到抑制。表明 p5 3基因对由于石英尘作用后的 AM培养上清所致 2 BS的非正常增殖和胶原合成有抑制作用 ,而对正常 2 BS无明显影响。
The influence of wild type p53 gene on the effects of surfactant from dust exposed AM on the abnormal proliferation and function of fibroblast(2BS).After the addition to 2BS of surfactant from macrophge exposed to dust both in vivo and in vitro for 24 hours.2BS was transfered by foreign wild type p53 gene with lipofection and remained in culture for 24 hours.Results:In comparison with non transfered group,the cell number(MTT) and the collagen content were significantly increased in transfered group.Those fidings suggested that the p53 gene might inhibite the abnormal proliferation and collagen synthesis of 2BS which were induced by surfactant from quartz exposed macrophage and not influence the normal 2BS.
