肌苷苦味酸速率测定方法学探讨

摘要 目的 :从反应原理、反应历程研究不同干扰物对肌苷 (Cr)苦味酸 (P)速率测定的干扰 ,并作定量描述。方法 :监测各干扰物与 P的反应历程曲线 ,分别检测各系列浓度干扰物对纯 Cr、血清 Cr的干扰。结果 :葡萄糖、丙酮、 α-酮戊二酸、抗坏血酸、头孢唑林钠、多巴胺、血红蛋白在病理、用药浓度、最大干扰值分别为 8.7%、 18.4%、 5 3%、 10 3%、2 0 %、 11%、 - 2 4%。高浓度 P与 Na OH试剂 ,延迟时间 2 0秒 ,测定时间 (2 0~ 40 )秒可基本消除生理浓度快反应干扰物的影响。同步反应干扰物的影响可询问病情、避开药物残留期、血清加标回收的办法予以消除。 Objective:The studies for the mechanism and the course of the interference to kinetic serum creatinine measurement were done to quatitative describe.Method:We continuous monitor the reactions course of the interference substances with the alkaline picrate,the value of the interference to the pure creatinine and the serum creatinine were independently determined.Result:In some mobid、drug concentration,the largest interference value of glucose、acetone、α ketoglutaric、ascorbic acid、cefazoline sodium、dopmine、hemoglobin were 8 7%、18 4%、53%、103%、20%、11%、-24%.The interference caused by the fast reaction substances at physiological concentration can be eliminated by the high concentration of picrate and NaOH reagent,and the use of 20 seconds of delay time and (20~40) seconds of measuring time.For the interference of the synchronous reaction substances,we may inquire the patients situation,avoid the drugs residual period,or standard addition method to decrease the interference.
