摘要 应用高效液相色谱-共振瑞利散射联用技术建立了测定依替米星的新方法。采用滂胺天蓝作为分子探针,共振瑞利散射(RRS)检测波长设为λex=λem=361nm,通过优化分子探针浓度、分子探针流速、柱后管长和pH等影响因素,确定了最佳实验条件,在该条件下依替米星的检出限(信噪比为3)为1.0mg/L。以妥布米星为内标物,以内标法定量,依替米星与妥布米星的峰面积比与依替米星的质量浓度之间的线性响应范围为2.5~60mg/L。将该方法应用于大鼠血清中依替米星的药代动力学研究,结果满意。 A method of high performance liquid chromatography-resonance rayleigh scattering(HPLCRRS)had been developed for the determination of etimicin by using pontamine sky blue as spectroscopic probes.The RRS detection wavelength was set atλex=λem=361nm.The parameters of the experiment condition and post column-probe solution on the sensitivity have been optimized.Under the optimal experiment conditions,the limit of detection for the etimicin was 1.0mg/L(S/N=3).With the internal standard(IS)of tobramycin,a linear calibration curve ranged from 2.5mg/L to 60mg/L was obtained.The proposed method was applied for the determination of etimicin in rat serum and proved to be suitable for the pharmacokinetics study of etimicin in rat following intramuscular injection.
