摘要
采用苯酚-硫酸法建立了灵芝多糖的荧光光谱测定方法。当葡萄糖浓度为20~100μg.mL-1时,λem=518nm处荧光强度与葡萄糖浓度有良好的线性关系,相关系数为0.9994,检出限为6.35μg.mL-1。用拟定的方法对9种灵芝样品进行了测定,其多糖含量在0.712%~2.548%之间,与《药典》法测定结果一致。加标平均回收率为99.82%,相对标准偏差为3.46%。该法结果稳定、重复性好,可用于天然产物中多糖含量的分析。
A method of fluorescent spectrometry by using the phenol-sulfuric acid was applied to determine the ganoderma polysaccharide.There is a good linear relationship between the fluorescence intensity and the content of ganoderma polysaccharide in the range of 20to 100μg.mL-1 atλem= 518 nm,and correlation conefficient is 0.9994,detection limit is 6.35μg.mL-1.Nine kinds of ganoderma lucidums were tested,and the ganoderma polysaccharide content was between 0.712% and 2.548%,which was consistent to the results of Pharmacopoeia.The average recovery is 99.82%,RSD is 3.46%.The method has good stability and repeatability,and can be used for the determination of polysaccharide content in natural products.
