摘要
根据Shciner提出的房室模型,设计了一种计算机程序,用于药代动力学与药效动力学结合模型效应参数的计算。本程序采用一种随机模式搜索寻优的算法,通过对给药后血药-时间和效应-时间的数据同时拟合,估算房室模型的效应参数。这种程序上机后,可得到最小方差的最佳估算值。它可以分别对静注、肌注和滴注三种给药方式进行效应参数的估算。本文以二房室静注模型为例,对呋喃苯胺酸的药代动力学与药效动力学结合模型的曲线拟合和效应参数进行估算。
A computer-program used in a compartment model approach proposed previously by Sheiner to the simultanous characterization of the pharmacokinetics and pharmacodynamics of a drug was designed. A randomized pattern search method to estimate the compartment parameters was adopted, in simultaneously curve-fitting plasma concentration-time data and effect data following the drug administration. The optimal values could be rapidly selected to get the minimal square error by computer. The computer-program has been applied to simultaneous fitting of pharmacokinetics and pharmacodynamics of furosemide according to two compartment model and used to estimate the parameters of the model.
