摘要
[目的]研究Y染色体微卫星基因座DYS441,DYS442在吉林省朝鲜族和汉族男性人群中的分布特征.[方法]采用PCR扩增结合聚丙烯酰胺凝胶电泳技术和银染显带方法对205名朝鲜族男性和207名汉族男性DYS441,DYS442基因座进行检测,并用Arlequin 2000软件进行数据分析.[结果]在朝鲜族男性人群DYS441基因座中检测到8个等位基因,个人识别能力为0.749 3,DYS442基因座中检测到7种等位基因,个人识别能力为0.508 3;在汉族男性人群DYS441基因座中检测到8个等位基因,个人识别能力为0.803 1,DYS442基因座中检测到5种等位基因,个人识别能力为0.627 3.分析由2个基因座共同构成的单倍型,在朝鲜族人群中检测到23种,在汉族人群中为31种,其单倍型变异度分别为0.874 6和0.927 2.[结论]DYS441和DYS442基因座具有较高的遗传多态性,是较理想的遗传标志,可为朝鲜族和汉族人群法医学个人识别、亲权鉴定及父系遗传学研究提供基础资料.
OBJECTIVE To study the distribution of microsatellite DYS441,DYS442 in Y chromosome from Korean-Chinese and Han nationality in men,Jilin Province.METHODS The polymorphisms of DYS441,DYS442 from 205 of Korean-Chinese and 207 of Han nationality in men were detected by multiplex polymerase chain reaction(PCR) and technique of polyacrylamide gel electrophoresis and silver staining.The data were analyzed with Arlequin 2000.RESULTS Total eight alleles of genetic DYS441 were observed and the calculated discrimination power was 0.749 3 while total seven alleles of genetic DYS442 were observed and the calculated discrimination power was 0.508 3 among Korean-Chinese males samples.In all eight alleles of genetic DYS441 were observed and the calculated discrimination power was 0.803 1,five alleles of genetic DYS442 were observed and the calculated discrimination power was 0.627 3 among Han males samples.Haplotypes,which constitute by two locus together,were analysed,indicating that tne haplotypes were found 23 in Korean-Chinese and 31 in Han nationality.Gene diversity of haplotype was 0.874 6 and 0.927 2,respectively.CONCLUSION DYS441,DYS442 loci in this study were found to have hight heterozygsity and polymorphic information content.Consequently they could provide useful markers for genetic purposes.The results also indicate valuable data for Korean-Chinese and Han nationality group genetic database and play an important role in Chinese population genetic application.
