兴安白头翁常春藤皂苷元的含量测定

Determination of content of hederagenin in the Pulsatilla dahurica(Fisch.) spreng

[目的]建立兴安白头翁药材中常春藤皂苷元含量的测定方法.[方法]采用反相高效液相色谱法,色谱柱为BDS HYPERSIL C18色谱柱(4.6mm×250.0mm,5μm),流动相为乙腈∶水(85∶15),流动速度为0.5mL/min,柱温为30℃,检测波长为210nm.[结果]常春藤皂苷元的线性范围为1～100mg/L(r=0.9995),平均加样回收率为107.86%,RSD为0.78%.[结论]反相高效液相色谱法准确,专属性强,可作为兴安白头翁的含量测定方法.

OBJECTIVE To develop a determination method for hederagenin in the Pulsatilla dahurica（Fisch.） spreng.METHODS Hederagenin was determined by RP-HPLC method.The RP-HPLC profiles were determined at 30℃ on BDS HYPERSIL C18 column（4.6 mm×250.0 mm,5 μm）.The mobile phase was acetonitrile water（V∶V=85∶15） at a flow rate of 0.5 mL/min.The detection wavelength was set at 210 nm.RESULTS The calibration curve was linear in the range of 1~100 mg/L（r=0.999 5）,the average recovery was 107.86% and RSD was 0.78%.CONCLUSION RP-HPLC method is accurate and specific as a determination method for the Pulsatilla dahurica（Fisch.） spreng.
