摘要 对浙江贝母组织培养材料进行了染色体显微鉴定技术和方法的研究。通过对组培材料的取材时间、预处理药剂的选择、浓度、时间、离析方法、离析药剂、浓度和时间以及不同取材部位(根尖、愈伤组织、芽尖)的系统试验获得了较佳制片效果,表明本染色体显微鉴定技术能较好地用于贝母离体组织培养材料的鉴定。 The method and technique in chromosome determination of cultured material of Fritillaria thunbergii were studied. A series of experiments were conducted with a view to obtaining good separation of chromosome for microscopie determination: such as the experiments on the time of cutting material, the selection of pretreatment chemicals, concentration and time of pretreatment, the selection of hydrolytic compounds, hydrolyzing time and the comparison of different cultured material. Based on the results of experiment, a good method and process was developed, which appears to be most successful with cultured material of Fritillaria thunbergii in the determination of chromosome.
