摘要
介绍新一代噻唑烷2,4二酮类治疗Ⅱ型糖尿病的第一个新药曲格列酮。着重对曲格列酮的药效学、临床治疗以及毒副作用等方面作简要综述。曲格列酮对非胰岛素依赖性糖尿病具有独特的药效,该药可提高胰岛素的作用,不刺激胰岛素分泌,降低血糖,但不引起低血糖,不增加体重。特异体质的患者服用曲格列酮会引起肝脏毒副作用,但目前尚不能认为易感亚群患者服用曲格列酮能引起肝脏毒副作用。曲格列酮是一个治疗非胰岛素依赖性糖尿病的新型胰岛素增敏剂,值得对其在非胰岛素依赖性糖尿病的治疗方面进一步进行临床研究,以便推广应用。
Troglitazone (TGZ)isthefirstoneinnewclassoforalantidiabeticagentswhichbecome availableforthetreatmentofpatientswithNIDDM .TGZ ,itspharmacodynamics ,clinicaltrialsand toxicologywerereviewed .TGZhasspecificmechanismofactionfordecreasinginsulinresistance .It improvesinsulinactionbutdoesnotstimulateinsulinrelease ,andreduceshyperglycaemiawithoutglu cose loweringresponse .TGZdosenotincreasebodyweightofpatients .Liverinjuryinidiosyncratic causedbyTGZwasstudied .However ,itisnotsurewhetherthisagentisharmfultothehealthinthe subgroupofsusceptiblepatients .TGZisanoveloralinsulin sensitizingagentfortreatmentofNID DM .ItisworthytostudyitfurtherfortreatmentofpatientswithNIDDM .
