摘要
目的 探讨高胆固醇血症作用下 ,兔胆管胆道口括约肌动力功能变化及其与一氧化氮合酶间的关系 .方法 纯种新西兰兔 1 6只 ,随机分为 2组 ,每组 8只 ,对照组饲以标准饲料 ,实验组饲以标准饲料加胆固醇 ,每日 1 g,每周喂 6 d,停用1 d,共 4wk.采用灌注式测压法测定并记录胆道口括约肌压力曲线 ,图像分析仪对胆道口括约肌中一氧化氮合酶含量进行分析 .结果 实验组和对照组胆道口括约肌低压壶腹段基础压分别为 2 .79k Pa± 0 .81 k Pa和 1 .5 6 k Pa± 0 .37k Pa,最大收缩波幅分别为 0 .85 k Pa± 0 .6 0 k Pa和 2 .41 k Pa± 0 .79k Pa,两组间差异显著 (P<0 .0 1 ) .胆道口括约肌高压带的基础压分别为 1 8.45 k Pa± 6 .0 7k Pa和 9.2 4k Pa± 1 .31 k Pa,最大收缩波幅分别为 0 .5 6 k Pa± 0 .2 9k Pa和 1 .1 6 k Pa± 0 .45k Pa,两组间差异十分显著 (P<0 .0 1 ) .一氧化氮合酶总数两组间无明显差异 .结论 高胆固醇血症可诱导兔胆管胆道口括约肌动力功能失调 ,主要特征为胆道口括约肌处于挛缩状态 .原因可能与高胆固醇改变了细胞膜的生物学特性致胆道口括约肌对一氧化氮的反应异常或者与降低。
AIM To investigate the relationship between the motility of Oddi′s sphincter and its quantity of nitric oxide synthase in rabbits with hypercholesterolemia. METHODS16 New Zealand rabbits were divided into two groups and fed for 4 weeks: control group ( n =8), normal diet and experimental group ( n =8), normal diet and cholesterol. Perfusion manometry was performed, pressure curve of sphincter of Oddi was recorded and quantity of nitric oxide synthase was analyzed. RESULTS The basal pressure of the two groups were (2.79±0.81)kPa and (1.56±0.37)kPa, and the amplitude of phasic contraction were (0.85±0.60)kPa and (2.41±0.79)kPa in low pressure ampulla of Oddi′s sphincter, respectively. There was significant difference between the experimental group and control group ( P <0.01). The basal pressure of two groups were (18.45±6.07)kPa and (9.24±1.31)kPa and the amplitude of phasic contraction were (0.56±0.29)kPa and (1.16±0.45)kPa in high pressure zone of Oddi′s sphincter, respectively. There was significant difference between the experimental group and the control group ( P <0.01). There was no significant difference in total quantity of nitric oxide synthase between the experimental group and the control group. CONCLUSION The diastolic function of sphincter of Oddi is impaired by hypercholesterolemia and the sphincter of Oddi is in a spasmic state, which may be caused by the hypercholesterolemia related abnormal response to nitric oxide or the decreased activity of nitric oxide.
