摘要
目的 研究巨噬细胞移动抑制因子(MIF)及Th1/Th2型细胞因子在原因不明早期复发性流产(UERSA)患者血清和蜕膜中的表达及意义.方法 选取因原因不明早期复发性流产患者22例(UERSA组)及同孕龄正常早孕妇女25例(正常早孕组),检测两组外周血清中和蜕膜中MIF、IL-2和IL-10的浓度.并分析血清和蜕膜组织中MIF水平与IL-2水平的相关性.结果 两组血清和蜕膜中均有MIF和IL-2、IL-10的表达.UERSA组血清、蜕膜组织中MIF、IL-2、IL-10分别为(23.4±6.9)mg/L、(65.6±20.4)ng/L、(73.5±32.1)ng/L和(20.6±6.1)mg/L、(42.6±17.6)ng/L、(47.6±17.1)ng/L,正常早孕组分别为(38.4±9.8)mg/L、(39.3±12.4) ng/L、(93.4±25.4)ng/L和(33.7±8.6)mg/L、(27.2±9.5)ng/L、(64.7±28.2)ng/L;血清和蜕膜中MIF和IL-10 UERSA组均低于正常早孕组;IL-2水平则明显高于正常早孕组,差异有统计学意义;血清和蜕膜组织中MIF水平与IL-2水平呈负相关(r=-0.78、-0.65).结论 妊娠期母胎界面MIF表达降低和Th1/Th2型细胞因子比率失衡,可能在原因不明早期复发性流产的发生发展中起重要作用.
Objective To explore the expression of macrophage migration inhibitory factor (MIF) and Th1/Th2 cytokines in serum and deeiduas in women with unexplained early recurrent spontaneous abortion (UERSA). Methods 22 women with unexplained early recurrent spontaneous abortion ( UERSA group) and 25 women with normal early pregnancy ( normal pregnant group) were recruited from Heze Municipal Hospital between August 2010 to May 2011. Th1/Th2 cytokines were represented by IL-2/IL-10. Levels of MIF, IL-2, IL-10 in serum and deciduas were measured by enzyme linked immunosorbent assay ( ELISA). The relationship between the levels of MIF and IL-2 in serum and dociduas were analyzed in the UERSA group and normal pregnant group. Results MIF, IL-2 and IL-10 were expressed in serum and deciduas in the UERSA group and the normal pregnant group. In the UERSA group, the expression of MIF, IL-2, IL- 10 N/S was (23.4 ± 6.9) mg/L, (65.6 ± 20.4) ng/L, and (73.5 ±32.1 ) ng/L, respectively, in serum and (20.6±6.1 ) mg/L, (42.6±17.6) ng/L, and (47.6±17.1 ) ng/L, respectively, in deeiduas, while in the normal pregnant group, they were (38.4±9.8) mg/L, (39.3 ±12.4) ng/L, and (93.4 ±25.4) ng/L, respectively, in serum and (33.7 ±8.6) mg/L, (27.2 ±9.5) ng/L, and (64.7 ±28.2) rig/L, respectively, in deeiduas. Serum and decidua MIF and IL-10 levels were markedly lower in the UERSA group than those in the normal pregnant group, while IL-2 levels were significantly higher in the UERSA group. There was a negative correlation between MIF and IL-2 in serum ( r = -0.78 ) and in meeiduas ( r = -0.65 ). Conclusion Lower levels of MIF and the imbalance of Th1/Th2 in serum and deeiduas may contribute to the occurrence of unexplained early recurrent spontaneous abortion.
