摘要
目的:研究黄芩对鱼藤酮所致帕金森(PD)模型大鼠的保护作用。方法:Wistar大鼠每日颈、背部皮下注射鱼藤酮葵花油乳化液制备PD大鼠模型;阳性组及不同剂量药物组在每日注射鱼藤酮之前1小时灌胃给药。ELISA法检测大鼠脑硫氧还蛋白(Trx);荧光探针法检测纹状体活性氧(ROS);分光光度法检测纹状体谷胱甘肽(GSH)、黑质丙二醛(MDA)含量。结果:造模后模型组大鼠出现一定程度的行为学变化,模型组Trx含量明显低于空白组、阳性组和低剂量组,药物可以一定程度上抑制氧化应激反应。结论:黄芩可以提高帕金森大鼠脑内Trx的表达,对鱼藤酮所致帕金森模型大鼠具有保护作用,这种作用可能与Trx相关。
Objective: To study the protective effect of Scutellaria on rotenone - induced Parkinson (PD) rats and its mechanism. Methods: Wistar rats subcutaneous injected daily with rotenone sunflower oil emulsion for 5 weeks for the prepa- ration of rat model of PD; rats of positive group and different doses groups were administered by i.g. daily 1 hour before hy- podermic injection, ELISA for detection of rat brain thioredoxin (Trx) ; fluorescence probe method for the striatum reactive oxygen species (ROS) ; spectrophotometric for glutathione ( GSH), malondialdehyde (MDA). Results:Rats in model group appeared some changed behavlor, thioredoxin in their brain were significantly lower than the control group, positive group and low dose group. The oxidative stress in rats were reduced by Scutellaria in some extent. Condusion:Scutellaria can improve expression of thioredoxin in rat' s brain, and has a protective effect on the rotenone - induced PD rats. This protective effect may be related to Trx.
