摘要
本文观察了绞股蓝总皂甙(gypenosides,GP)对大鼠实验性急性心肌缺血及培养心肌细胞缺糖缺氧损伤的保护作用。实验发现,大鼠在冠状动脉结扎后,早期腹腔注射绞股蓝总皂甙,可使缺血24h的心肌梗塞范围显著缩小;并使缺血6及10h的大鼠血清磷酸肌酸激酶(CPK)和乳酸脱氢酶(LDH)明显降低;使缺血后30min时缺血边缘区心肌的超微结构损伤明显减轻。体外实验表明,绞股蓝总皂甙能保护大鼠培养心肌细胞缺糖缺氧性损伤,抑制缺氧缺糖6h心肌细胞CPK及LDH的释放。证明绞股蓝总皂甙对缺血及缺氧心肌具有一定的保护作用。
The effects of gypenosides (G) on ischemic myo-cardium were studied in conscious animals and incultured cardiac cells from neonate rats. The resultsshowed that the administration of G to rabbits (25mg/kg, iv) and rats (50mg, ip) significantly decreased thesize of infarction induced by left coronary ligation.The ultrastructural damage of ischemic myocardiumin the treated group was less in rats,G dramaticallyreduced the ZST, NST, the number of Q wave afterthe coronary ligation in rabbits, and that G inhibitedthe increase of creatine phosphokinase (CPK), lactatedehydrogenase (LDH), free fatty acid and TXB_2 inblood after the ligation. 50~200μg/ml G significantlydepressed the release of CPK and LDH from culturedcariac cells during anoxia in a concentration-depen-dent manner. The experiment results suggest that Ghas a protective affects on ischemic and anoxiamyocardium.
