酸甘生津方及其拆方对干燥综合征模型小鼠颌下腺BAFF的影响

Impacts of Suangan Shengjin Formula and its Decomposed Recipes on BAFF of the Salivary Glands for the Model Mice with Sjogren's Syndrome

目的观察酸甘生津方及其拆方对干燥综合征(SS)模型小鼠颌下腺组织的病理表现及免疫组化中B细胞活化因子(BAFF)的影响。方法将60只BALB/C小鼠随机分为全方组,养阴、活血、解毒3个拆方组及空白组,模型组共六组。除空白组外,其余五组小鼠采用同种小鼠颌下腺接种方法造模,造模2周后用免疫组化法观察各组小鼠颌下腺BAFF含量的变化,分析比较各组间表达差异。结果组织切片总阳性率:全方组与活血组相等,与养阴组和解毒组比较,差异有统计学意义(P<0.05);SS模型小鼠颌下腺组织BAFF表达水平:各给药组均低于模型组,差异有统计学意义(P<0.01)。其中全方组、活血组及养阴组的BAFF表达水平均低于解毒组,差异有统计学意义(P<0.01),但此三组间比较差异无统计学意义(P>0.05)。结论酸甘生津方及其拆方对模型组小鼠颌下腺BAFF过度分泌有较好的抑制作用,可能通过调控BAFF表达来抑制B细胞异常活化。

Objective To observe the impacts of suangan shengjin formula and its decomposed recipes on the pathological manifestations of the salivary tissues and B cell activating factor belong to TNF family（BAFF）in immunohistochemistry（HIC）for the model mice with Sjogren’s syndrome（SS）.Methods 60 BALB/C mice were randomized into a whole-formula group,three decomposed-recipe groups for nourishing yin,activating blood circulation and detoxification separately,a blank group and a model group.Except the mice in the blank group,the mice in the other five groups received the allogenic salivary vaccination for modeling.In 2 weeks of modeling,HIC method was adopted to observe the change in BAFF content of the salivary glands for the mice in each group and analyze the expression differences among the groups.Results Concerning the total positive rate of the tissue slices：the rate in the whole-formula group was same as that in the decomposed recipe group for activating blood circulation.Compared with the decomposed recipe groups for nourishing yin and detoxification,the differences were significant statistically（P0.05）.Conclusion Suangan shengjin formula and the decomposed recipes can satisfactorily inhibit the excessive secretion of the salivary BAFF for the mice in the model group,which is probably due to the inhibition of B cell abnormal activation via regulating and controlling BAFF expression.
