大木枣休眠枝水培芽及其离体培养芽诱导的研究

Study of the Water Culture Bads and the Vitro Bads on the Dormant Branchs of Zizyphus Jujuba Mill Var.DaMuZao in Vitro Culture

:大木枣茎段离体培养中 ,早春休眠枝沙藏法是保存和延长取材时限的有效方法 ;将休眠枝置于温室 (温度 2 5～ 30℃ ,相对湿度 6 0 %～ 75 % )水培 ,取水培芽作外植体 ;外植体消毒的优选方法是 :洗洁净水清洗→ 75 %乙醇 10 s→饱合漂白粉上清液+吐温 - 2 0混合液 2 0 min;初代培养的适宜培养基为 MS附加 6 - BA 1.0 mg/l,IBA0 .1mg/l.这些结果可能对枣属植物的组织培养具有一定的参考价值 .

Result has showed thatas to in vitro tissue culture of stem in Zizyphus jujuba Mill Var· Da Mu Zao the method conserve early spring dormantbranch in stratification is efficient to conserve and prolong the time for taking the material.With the dormant branches put in greenhouse(indoor temperature is2 5～ 30℃ ,relative humidity is the6 0 %～ 75 % ) sprouted in water culture,then taking the water culture bads as the explant. The best method of surface sterilization is washing it with detergent(what is the detergent of the concentration used)→ 75 % alcohol(10 seconds)→ saturation solution of bleaching powder and twenn2 0 mixture solution(2 0 minutes) .The optimal elementary generation incubation is MS added6 - BA 1.0 mg/l,IBA 0 .1mg/l. These resultsare generaly useful in tissue culture of zizyphus jujuba mill.
