波动和恒定的葡萄糖浓度对大鼠肾小球系膜细胞TRPC6和TRPC1表达的影响 被引量:4

摘要 目的研究波动和恒定的葡萄糖浓度对体外培养的大鼠肾小球系膜细胞(GMC)瞬时受体势TR-PC6和TRPC1表达的影响,探讨糖尿病肾病的发病机制。方法将体外培养的各组系膜细胞分别在波动和恒定葡萄糖浓度中分别培养24、48、72 h后,进行细胞学形态结构观察,MTT法测定细胞存活率,RT-PCR检测细胞TRPC6和TRPC1特异性条带灰度的表达情况。结果波动血糖组在TRPC6基因的表达较恒定高糖组差异有统计学意义(P<0.01)。结论波动葡萄糖浓度对GMC的损伤以及促进细胞凋亡方面较恒定葡糖糖浓度更严重,是导致糖尿病肾病发生和发展的原因之一。
