大肠菌群快速检测纸片配方与工艺研究

Study on the Formula and Technology of the Rapid Paper Method of Coliform Bacteria Examination

将大肠菌液制成已知准确浓度的模拟菌液作为试验样品,通过对影响自制大肠菌群快速纸片效果的配方和工艺参数进行研究。筛选出纸片配方为:乳糖2.5g、蛋白胨1.5g、胆盐0.15g、TTC(4%)2mL、琼脂0.1g、NaCl0.3g、K2HPO34g、溴甲酚紫2mg、乳化剂S(1%)2mL、pH6.8～7.0;筛选出工艺参数为:灭菌温度110℃、灭菌时间15min、浸液时间1min、浸液温度60℃、烘干温度55℃。通过重复性和准确性试验,证明自制纸片和发酵法检测的结果总符合率为94%,经统计学处理P>0.05,说明两法的检测结果无显著区别。

The accurate concentration of analogy bacterial liquid was made as test samples using coliform bacteria liquid.And study the formula that will influence the effect of self-made rapid paper and the processing parameters.The paper formula：Lactose bile salt 2.5g,peptone 1.5g,bile salts 0.15g,TTC（4%）2mL,agar 0.1g,NaCl 0.3g,K2HPO3 4g,bromocresol purple 2 mg,emulsifying agents S（1%）2mL,pH 6.8～7.0.The processing parameters：sterilization temperature110℃、sterilization time 15min、immersion time 1min、fluid temperature 60℃、drying temperature 55℃.The Self-made paper for total coincidence rate of detection result was 94%.The test result was no significant difference（P0.05）.
