摘要
目的探讨八肽胆囊收缩素(CCK-8)治疗类风湿关节炎(RA)的可行性及机制。方法 DBA/1J小鼠60只,随机分为CCK-8高、中、低剂量组和对照组各15只。将CII完全乳化液于小鼠尾根部皮内多点注射(100μl/只),第21天再次同剂量追加免疫一次,建立CIA模型。于第二次增强免疫同时,对照组腹腔注射生理盐水100μl/只;CCK-8高、中、低剂量组腹腔分别注射CCK-8 1、5、10 nmol/只,每天注射1次,连续注射7 d。采用酶联免疫反应法检测各组关节滑膜组织TNF-α和IL-6水平。结果 CCK-8各剂量组关节组织TNF-α和IL-6水平均明显低于对照组;中、高剂量组明显低于低剂量组(P均<0.05)。结论 CCK-8能降低CIA小鼠TNF-α和IL-6水平,此可能是其治疗RA的机制之一。
Objective To investigate the feasibility and mechanism of rheumatoid arthritis(RA) treated by CCK-8.Methods Sixty DBA/1J mice were divided randomly into CCK-8 high,medium low dosage groups and control group with 15 in each,then were immunized subcutaneously at the base of the tail with emulsified chicken CII and Freund’s complete adjuvant to set CIA model.The CCK-8 groups were treated with different doses of CCK-8,the control group was given normal saline.The levels of TNF-α and IL-6 in synovium of mice were detected by ELISA.Results The levels of TNF-α and IL-6 in synovium of CCK-8 groups were much lower than those in control group,which of the high,medium dosage groups were significantly lower than those of low dosage group(all P〈0.05).Conclusion CCK-8 can down-regulate the TNF-α and IL-6 levels in synovium of CIA mice,this maybe its therapeutic mechanism for RA.
